Adipotide is a peptidomimetic that fuses a prohibitin-targeting sequence to a pro-apoptotic domain, designed to trigger apoptosis in the vasculature of white adipose tissue. The famous weight-loss data are from obese rhesus monkeys, published in Science Translational Medicine in November 2011.
Arrowhead took it into a first-in-human phase 1 trial not in obesity but in men with castrate-resistant prostate cancer and no standard treatment options, with the secondary question of whether reducing white fat could slow tumour growth. The design was classical 3+3: up to five dose levels, three patients each, escalating on observed safety.
Dosing was by subcutaneous injection once daily for a single 28-day cycle, and the trial's stated objectives were maximum tolerated dose, pharmacokinetics and mechanistic markers. Crucially, neither the sponsor announcement nor the trade coverage discloses the milligram or mg/kg amounts at any dose level.
