What the literature reports

MOTS-c is a 16-amino-acid peptide encoded not by nuclear DNA but by a short open reading frame within the mitochondrial 12S rRNA gene. It is one of the founding members of the mitochondrial-derived peptide family.

Cellaire classifies MOTS-c as preclinical evidence. The founding Lee et al. paper (Cell Metabolism 2015) established the 51-bp sORF, cytoplasmic translation, conservation across 14 species, and detection of the peptide in rodent tissues and human plasma. Reported metabolic and insulin-sensitizing effects come almost entirely from mouse and rat models.

Molecular overview

MOTS-c (Mitochondrial Open-reading-frame of the Twelve S rRNA type-c) is the peptide MRWQEMGYIFYPRKLR, translated from a 51-base-pair sORF within the mitochondrial 12S ribosomal RNA gene.

It is the first well-characterized mitochondrial-derived peptide with metabolic activity in mammals. Circulating levels vary with exercise: skeletal-muscle MOTS-c rises roughly 11.9-fold after acute exercise in humans.

Preclinical work reports AMPK activation, insulin sensitization, glucose disposal in muscle, and anti-inflammatory activity in mouse models of obesity, diabetes, and aging.

MOTS-c is not FDA-approved for any indication and is on the FDA Category 2 bulk substances review list.
